During the course, participants will learn theoretical and practical aspects of sample preparation and data collection for cryo-EM
What is CryoEM ? – EM vs structural biology : strategies and objectives.
Single particle analysis: quality control by negative staining, vitrification of macromolecules
High Pressure Freezing: vitrification of cells and tissues
Data acquisition: low dose / automatic data collection.
How each machine works: HPF, Glow discharge, Vitrobot.
At the end of the workshop, the student will be able to freeze one grid, to load a sample to the cryo-holder and then to microscope, and to do a small data collection.
