Students will use the state-of-the art equipment of the IBiSA high-throughput protein purification facility of the AFMB (Tecan robotics and microfuidics detection) on their own plasmids (up to 12/student) to optimize the production and purification of their recalcitrant proteins of interest in 96 well format. They will learn the protocols and the strategies and know how to transfer them to a laboratory equipped with standard equipment.
The AFMB facility can perform all the steps from gene cloning to purified protein samples in high-throughput format. This service uses innovative high-throughput strategies and methodologies that have been mostly developed in-house and validated in AFMB by the purification of more than 10.000 proteins for customers, collaborators and big scale projects (twelve ANR and twelve EU grants). The staff of the facility has been teaching these protocols since 2010 to Polytech students and to >100 EU phD students through EMBO practical courses.
Students prerequisites
At the end of the workshop, the student will be able to verify the production and purification of his samples and be able to transfer HTP purification in 24/96 well format from E. coli cultures in their lab for further project. They will know how to design alone the best possible routes to have the maximum chances to get their proteins purified in big quantities as quickly as possible.
