The aims of the practicals are:

  • Learn the principle of NMR and its applications in structural biology
  • How to prepare samples for NMR studies
  • How to analyze NMR spectra to get structural information on protein samples

Timeline

1. A few weeks before the first meeting, all participants will be asked to complete a form including their contact details, their laboratory, their thesis topic, the methods used “routinely” and the complementary skills sought in this workshop. Doctoral students need to inform their thesis supervisor of their approach (their approval is mandatory).

Day 1
– Initiation to NMR: definition and foundations, techniques and examples of application Lecture.
– Introduction to the equipment.
– Sample preparation for liquid NMR and acquisition of 1D spectrum

Day 2
– Introduction to homonuclear and heteronuclear 2D NMR course.
– Acquisition of a 2D homonuclear spectrum 1H-1H.

Day 3
– Acquisition of a 2D heteronuclear NMR spectrum 1H-15N.
– Analysis of the folding state of a protein.
– Interaction studies protein-protein and protein-ligand complexes.

Objectives

At the end of the workshop, the student will be able to prepare samples for NMR studies and to analyze NMR spectra to get structural information on protein samples.
