The Proteomics Platform of the Institut de Microbiologie de la Méditerranée is part of the Marseille Proteomics Platform (MaP) network, supported by the IMM, La Région, IBiSA, CNRS & AMU; the platform is equipped with a protein sequencer and three mass spectrometers, one of which is a high-resolution spectrometer coupled to a nano-chromatography chain, and another coupled to a proton/deuterium exchange module. The platform is involved in the physico-chemical characterisation of proteins from homogeneous solutions to whole proteomes containing thousands of proteins. It uses Edman sequencing techniques to characterise the primary N-terminal structure, and mass spectrometry to identify proteins by their peptide fingerprint after enzymatic digestion and MS & MS/MS (bottom-up proteomics) analyses. Amino acids targeted by chemical or post-translational modifications (phosphorylation, glutathionylation, acylation, glycosylation, sulphuration, etc.) are also finely characterised. The platform also uses tools to study the conformational dynamics of proteins, by mass spectrometry and H/D exchanges, to shed light on their dynamic or fixed structural regions during protein-protein/protein-ligand interactions or under various physicochemical conditions.

Protein Sequencer/Mass Spectrometer: MALDI-ToF (MicroflexII, Bruker)/ESI-Q-ToF(SynaptG1, Waters) coupled with Acquity chromatography (Waters) and HDX/ ESI-Q-Orbitrap module (Q-Exactive plus, Thermofisher) coupled with Ultimate 3000 nanochromatography (Thermofisher).
